B-cells maturation

Dear everyone,

I have isolated spleen cells of mouse to get B-Cells that contain mRNA of target protein. Is it necessary to culture the cells (spleen cells) before B-cells isolation to get mature B cells with large amount of mRNA? Does anyone can help me, please.
