I urgently need a protocol that works well for protein extraction from gram-positive bacteria.  I am particularly looking to extract membrane and cytoplasmic fractions, which I intend to separate on  SDS-PAGE. Any suggestion on protocol information will be most appreciated.

There are many permutations of protein extraction buffers and processes for any organism. A good start would be Protein Purification by Scopes.
