nanoparticles delivery to the targeted cells

AsPA-RITC nanoparticles were synthesized . The concn of nanosuspension  maintained was 10 microgram/ml of cell solutions. The centrifugation was carried and cells were also washed with PBS for more than three times. FBS was  used as uptake buffer.The cells were incubated in hotwater bath at 37 for 15 and 30 min. When efforts were made to know whether the nanoparticles were internalized or not . No nanoparticles were found to go inside. I wanted to improve my experimental skills and would apperciate the suggestions of anybody having profound knowlege on this topic.
