I am a research associate and about to start with my project work. I would be working on gene synthesis so any guys who could help me with this would greatly appreciate. I have my protein sequences . What I need is to generate a  DNA sequence optimized for expression system of choice.

Which is the most  appropriate method for this- PCR assembly or LCR coupled to PCR ?

Also please let me know if there is any software available for oligo design?
