Dear. All
I am doing in situ hybridazation with whole mount Zebrafish embryo at Sheild Stage. however, the stain is so weak because the protein which I would like to see excist so littele amount at the stage. I am consideing using paraffin sectioning in situ method for the next step.
My question is which I should do first sectioning or staining. My concern is whether the embryo at that stage can stand sectioning because it looks weak. if anyone know it, could you give me suggestion?
