global mean normalization

Im new to qPCR based miroarray study and would really appreciate any advice.


I understand we need to use GeNorm vs glocal mean normalization for analysis of a large set of data.


GeNorm is used to identify stable the references genes from a set of genes in a give panel of samples.With the reference gene identified:


1. do we calculate mean Ct across all miRNAs per each sample separately

2.get global mean normalized values for each miRNA in given sample A.

3. we use the global mean normalized values each miRNA in given sample A and global mean normalized values of reference genes identified by Genorm to calculate delta delta CT method and difference in fold change?

Im confused between the roles of GeNorm and global mean normalization.
