Im new to methylation specific PCR (MSP)and would appreciate all the advice


I plan to do end-point MSP followed by gel electrophoresis.


I have a few doubts that I wanted to clear:


1. What is the gel loading control. Will it be actin as well?

2. I have also ordered control DNA (bilsulphite methylated/unmethylated DNA, and also non bisulphite converted unmethylated DNA). So I can check the specificity of my primers by using these control DNA right? If I do not get bands it means I need to re-design my primers?

3. I should also run a negative control (no template).
