I want to prove the cleavage of a plastidial mRNA of my interest. As I went through literature I have came across with RLM RACE as method to study sRNA mediated mRNA cleavages. But a 5’ cap or a free 5’ monophosphate is a prerequisite for this method. As far as I know plastid mRNAs doesn’t possess this. Can anyone suggest an alternative for this?
