SDS PAGE bands

Hi everyone,

 

I am wondering why the bands of the samples only looks like arched  down like that ( in the SDS PAGE ?

there are a number of possible reasons for this. most are constituents of the sample. 3 of the more likely culprits are lipids and/or salts and/or surfactants (other than normal concentrations of sds) in the sample.

 

sometimes the effect can be defeated by running the tracking dye to the very bottom or out of the gel but this may lead to loss of the smallest peptides from the gel.
