Hi everyone, first time poster here.

 

Usually in a luciferase assay you are putting the promoter of a certain gene on the luciferase gene. I'm trying to see if MIR34a expression is altered by a regulatory element/transcription factor binding sequence in its promoter. Can I use the microRNA host gene promoter in a luciferase assay? I have not seen this done with ncRNA in the literature, only mRNA. Please provide a reference if you know of one.
