does anybody know whether or not you need to do a counterstain when you're performing TUNEL????? i seem to remember from years ago that you do but the promega protocol doesn't mention it??

I would suggest a light counterstain using either hematoxylin or methyl green. Please check protocols below:

TUNEL Protocol

Gill's Hematoxylin Solution

Methyl Green Solution

Note LIGHT counterstain. If over do it, positive cells well be covered.

Good luck.

I think you need to do a counter stain. You could refer to the manual given by roche-applied for their tunel kits.

All the best
