I'd be grateful if anyone can help with this question. We have a very severe EAE protocol in our lab, where differences between wild type and knock-out strains of mice can be minimal due to the severity of the disease. So we induced EAE with a less severe protocol in our last experiment and, if anything, the mice developed MORE severe EAE instead of milder disease. Has anyone seen this before? If so, what is it due to? I should say that the only difference between this experiment and the one before it was that the CFA was freshly made for this experiment (oh and no, there were no mistakes with the dilution of reagents nor the injections - we triple checked as we did them).

Many thanks in advance.
