I am working on determining total phenolic compounds in molasses. A standard garlic acid curve was done and an equation was generated. And the products' gallic acid was similarly generated. The absorbance values of the tested product fell within the standard curve values. How do i use equation generated in the standard gallic acid curve and apply it with the aborbance values obtained with the product? Then how shall I calculate the phenolic acids in the product ? Your assistance is greatly appreciated.

You do this just the same as you would for say a BCA assay or any standard curve - you take the absorbance and put it into the equation generated by the standards and solve to give you concentration. Just be aware of the usual precautions - don't extrapolate; dilute unknowns so that the absorbances lie within the range the standard curve. Make sure you are fitting the right sort of curve. Many graphing programs will do the calculations for you if needed.

 

To calculate the concentration of the phenolics, you would need to the proportion of phenolics to other organics, then work from there.
