I wanted to quantify the levels of IL-2 in spleen cells.  Therefore I performed an ELISA, but the results were not as I expected, the non-stimulated cells had higher levels of IL-2 than the stimulated ones.

My question is, could be another reason, besides a problem with the stimulation solution, that caused this effect on the stimulated cells?

Could be a problem with the lysis buffer?
