i have to do invitro of dietary fiber, my sample is wheat bran, if any one have knowledge related to this please guide me

No idea, however, dietary fiber is usually considered to be long chain, indigestible (but not insoluble) sugars. So you could dissolve the food in water/buffer, digest out the short chain sugars (e.g use amylase) then measure what is left (HPLC??)

amylase won't work if the fiber is cellulose, but cellulase should work.
