I am  using  primary thymocyte cell culture and I find a decrease in ROS in the normal cell population during the first 30 min. I am using DCFH - DA dye  ( 100 um) ,15 min prior to cell acquisition by flow cytometry for 30 min.
Please suggest an explanation for such a decrease. I expected  no change.
