Hi !

I want to extract DNA from alcohol preserved skin tissue from turtles.

1.     Is there any treatment needed to the tissue to remove alcohol prior to extraction  procedure?

2.     I have tried Qiagen DNeasyBlood and tissue kit. I put the small tissue pieces in 180μL ATL lysis buffer with 20 μLProteinase k , 56°C                         overnight, even 48 hours but it  does not lyse completely. How can I do complete lysis?

         I am attaching my protocol.Attached File  DNeasy-Blood--Tissue protocol.pdf   63.95KB   1428 downloads

Please  someone please help me on this. This is my first post

                                                                                                   With regards
