Hi,

 

Has anyone tried to transform tomato via Agrobacterium using basta (phosphinotricin) as selecting agent? I've been trying several times but nothing works. It looks like tomato is very sensitive to basta, thus I didn't get any regenerating tissues; I occasionally got some shoots but they were all "escapes".

 

I am also transforming tomato using a different selection (hygromycin) and it worked; I obtained a number of T0 which are PCR-positive, expressed the gene, and resistant to hygromycin.

 

Any comments, suggestions, will be really appreciated. Thank you.
