Really basic qs- Triplicate generation for milk urea assay?

When generating triplicates for milk urea quantification by assay using a multi-plate reader is it common practice to:


a) de-proteinate the sample in 3 seperate eppendorfs and pippette from each eppendorf into a single well or

b ) de-proteinate the sample in a single eppendorf and pipette this sample into 3 wells?


I have a little lab experience and would have performed option b ) but a collegue disagrees, neither of us have much experience.
