Hi everyone.


This is the first time I'm posting.  I am looking to permeabilize E. coli colonies on agar plates.  I've read a ton about various methods but most of them usually perform the process in liquid culture. 


I've been looking into Tris-EDTA or Sodium Hexametaphosphate; however, niether of these techniques work very well in nutrient media. Best method I've used thus far is lysozyme treating the plates, but this requires several mls of liquid to be place overtop plate.  This causes my colonies to streak and turn into snot intermingling the colonies. 


Anyone have any ideas??????
