Vertical Electrophoresis

Dear all,

I am new in molecular biology and vertical electrophoresis. I was using a vertical unit (10x10cm) with inner buffer only covering the electrode. According my colleague I have to put the inner buffer contacting the gel to have a net charge but it works only covering the eletrode with the inner buffer, I mean, the inner buffer donīt touchs the top of the gel.
Iīd like to know if somebody else do like me, because I canīt explain (just suppose) how the proteins migrated to the botton of the gel, remember that the positive pole is in the botton of the chamber and in contact with outter buffer.
