Anyone here know how to do experiment on nutrient surplus using jurkat cell culture and what does it really mean? Please advise. Thanks in advance.

I am intending to seed 0.125million cells/ml in 24 well plates cell insert. Then try to add different amount of media at different wells. So i fixed the amount of cell in the each well and vary the volume of media. The hypothesis that i am trying to test is if Jurkat cells proliferate more in the condition of surplus media. 

But my concern now is if i add more media to the well, am i like diluting the cells? So i am trying to ask if there are any proper ways of testing this hypothesis.
