I want to use CRISPR to introduce a single nucleotide change in my gene of interest. The cell line i am going to use is very difficult to transfect . I need to use lentivirus to transfect. There is some lenti plasimid available so this is not the problem. The problem is how I can introduce the template DNA into the cell at the same time so that I can introduce a specific nucleotide change? Thanks a lot
