Hi all,

Has anyone tried transforming bacteria with a mammalian cell lysate (which has a plasmid, transiently transfected) after 48-72 hrs post transfection.
If so how many cells would be needed to do so.

I basically want to transect mammalian cells with a cDNA library, then do a intra cellular reporter assay and bin these cells which are positive using FACS. Then can i retrieve back the plasmids from the cells that gave me a positive signal, by transforming bacteria with the cell lysate.
help in this regard will be of very useful,
thanks,

Bala
