In situ hybridisations

Hi All,

I was wondering whether anyone out there doing in situ hybridisations on tissue sections could let me know how much probe they use per section? I'm currently using DIG-labelled probes on mouse testis sections (about 5 um thick). How many ng or ug of probe do you guys use per section?

Hi Ros
In my lab, a hybridization solution containing 3ng of probe per microliter was used. You just cover up the section with this hybridization solution. We used flourescently labelled probes, you might need more for the DIG ones.
