Hi everyone

 

I am looking for a suitable promoter which can derive minimal expression of the selection marker gene (e.g neo) in mammalian cells. I know that  SV40 promoter is weaker than CMV but I need to reduce the expression level even more. so I am thinking maybe I can use SV40 minimal promoter. I don't have any experience with this promoter and I don't know any commercial vector which contains this promoter and also I am not sure which part of the promoter is the minimal part, so I was wondering maybe someone can help me with this.

Viral promoters tend to be quite strong, try looking for a mammalian promoter if you are using mammalian cell cultures for the expression.
