Hi,

I am trying to chrosomally HA tag a gene on the chromosome in E.coli. The gene is the second gene in a three-gene operon, so I don't think it will be good to insert a whole plasmid after the gene. Does anybody have any experience doing this? What kind of suicide vector I can use in E.coli and where can I get these vectors? Or is it possilbe to tag this gene using homologous recombination? Any suggestion will be appreciated! Thanks!
