Problems with BRCA1 and BRCA2 staining for Immunofluorescence

I have been trying to find the best technique to get BRCA1 and BRCA2 foci in immunofluorescence staining of cells but with no avail.


I am using the following antibodies:


Calbiochem mouse BRCA-2 (Ab-1) #OP95

Calbiochem mouse BRCA-1 (Ab-1) #OP92

Santa Cruz mouse BRCA-1 (D-9)


Having perused a multitude of literature and attempting different techniques – pre-permeabilisation and different fixing– PFA and MeOH/Ac, different concentrations etc

I have yet to recreate the wonderful images seen in all the other journals.


Does anyone have any advice or tips on which technique is best? Perhaps I should consider using a different antibody?
