Our lab does bone marrow-derived dendritic cell preps where we culture bone marrow leukocytes in RPMI + GM-CSF + IL4 + 5% serum + NEAA + 1mM glutamine + sodium pyruvate + HEPES + penn strep, aspirate granulocytes at day 4, replace with fresh media, and harvest on day 7.  Our yields are always extremely low; from 500 million bone marrow leukocytes we usually get about 50 million DCs [phenotyped via CD80, CD86, IAb, CD11c, and Gr-1].  Is this a typical outcome or are we doing something wrong?
