I want to perform a transient transfection in a human cell line and then a short drug selection (i.e. puro, G418 etc few days) to enrich for transfected cells. However, I want to prevent the plasmid from integrating. Which drug (i.e. G418, Puro, zeomycin, hygro, etc) causes the most and which ones cause the least amount of DNA breaks? I assume the drugs that cause the least will also result in less plasmid integration. Any information on this?
