Hi,  I'm looking for pyrF competent cells, or at the very least a pyrF strain with a genotype amenable to transformation and stable maintenance of a plasmid, but haven't been able to find anything commercially available or through ATCC.  I'm also wondering if the cerevisiae URA3 promoter has any low-level activity in E. coli.  Any thoughts?  (Briefly, I want to replace a cassette containing a yeast promoter-driven URA3 in vitro and screen for success in bacteria without having to put URA3 under a bacterial promoter if remotely possible.)  Thanks very much in advance!
