I am wondering why I cannot differentiate three populations when I do flow on PBMCs. I've read that I should be able to see; monocytes, granulocytes, and my lymphocytes: however, when I do my FSC vs SSC; I just get one large group of cells.

Any ideas?

Without knowing the details of how you proceeded, the first thing I thought of is whether the voltage settings are correct ?

Thanks for your respons. I agree, it is hard to say much with some background info. The problem is I am using a new machine (gallios by Beckman), and it is a whole bunch different from using the usual BD LSR II that I was used to.
