16S / NGS and Bacillus

Hi all

 

Not sure if this is the right forum area but I'll give it a try. I've been doing some community analysis work with V5-V7 regions of the 16S gene from the indoor environment. When I look at contact agar plates taken from the same location, the plates have a lot of Bacillus, however, very little if any of my amplicons from my 16S NGS library were identified as Bacillus

 

Does anyone know the reason for this please?
