Hi all,


I am actually looking for the ada levels in serum. But, I faced a problem regarding the wavelength ... According to the protocol ... http://www.worthingt.../ada/assay.html there should be a decrease in the absorbance at the 265nm. 


I tried different dilutions of serum, following the same conditions given in the protocol ... but there is no reduction in the absorbance ( reduction in absorbance should show the conversion of adenosine to inosine). 


Some articles have mentioned that adenosine gives absorbance at 235nm ... rather than 265nm ... 


please guide me as to which wavelength is appropriate to use to get the correct result.
