Hello,

 

I have been performing a positive cell separation using DynaBeads to separate out CD4+, CD8+ and CD14+ cells from fresh PBMCs. For both the CD4+ and CD8+ cell separation I achieve good purities of around 95-98%. For my CD14+, monocyte, separation the highest purity I have gotten is only 69% besides I struggle with getting high enough cell numbers. I only manage to separate out 1-2% CD14+ cells from my PBMCs. 

 

I have been reading up on alternative (and possibly easier) methods of monocyte separation but I was hoping someone has used the CD14+ DynaBead separation and has some advice on why I can't seem to get it to work.
