PMN isolation using histopaque

Hi everyone,


I have been attempting to isolated PMNs and PBMCs from human blood by layering histopaque 1077 onto 1119. However I fail to get to separate leukocyte bands. I am extremely slow and careful with my layering but I suspect the 2 density gradients are diffusing, does anyone have any suggestions?
