I have been performing DAB immunohistochemistry on mice brain sections; sometimes (but not always) when the gelatin-slide bound sections are put into the water bath for their final dehydration before coverslipping, the sections crumple together and sometimes detach. All sections are thoroughly dried in a fume hood, I also tested this using a warm water bath which made it worse. The next step is to try with a water bath on ice, though I've as yet only seen protocols using a warm water bath. For the time being I've switched to superforst slides which are working well, but it would be nice to have the option of using the gelatin coated slides.
does anyone have any ideas on what may be causing the sections to crumple and detach in the water bath?
