I am trying to clone 2 large human Ig subfamily proteins using the Gateway cloning system. I first started trying to express in a bacterial system and have recently moved to a mammalian system but have had no luck in either. Does anyone have any experiance using this system??? I have sequenced both expression vectors and they are correct. I have also used proteosome inhibitors in the mammalian expression.

Any help/advice would be much apreciated as I have been working on htis for over 7 months!
