Hi All,

I am starting a new experiment in my lab. I am using lung cytosol form mice and will expose them to different treatments for 4h. Then it  will be electrophoresed to resolve DNA-protein complexes.

I got the lungs from the animals and homogenized the tissue in ice cold 10mM Tris-HCL (PH 7.8) then centrifuged them at 10,000 xg for 20 min in 4oC.  The samples are now stored in -80oC.

How can I use the cytosol in vitro? I mean do put in culture plates and just the treatments? does it need any media for that?
