How do I induce the formation of beta amyloid plaques in SH-SY5Y cell line and h

Hello everybody! 

 

I am new in this field and I have a lot of questions! :b 

 

I would like to know if there is a specific protocol for the induction of beta amyloid plaques in SH-SY5Y cells. I read something about the use of amyloid beta peptide 1-42, but there was no information about quantity, time of incubation, analysis... 

 

I want to analyse if an interaction between two known proteins is capable of modulating the levels of beta amyloid aggregation in the cell.

 

About the analysis, I took a look on a kit that quantify the secreted APP and amyloid beta (ELISA), but is too expensive! Is WB enough to quantify the beta amyloid expression? Should I use specific Ac for Abeta 1-42 and 1-40 or only 1-42 is Ok?
