I need some help with these questions please...huh.png smile.png

 

1. List which of the following strains of E. coli would grow under the conditions given below.

 

 strain A hisA15

 strain B leu-258

 strain C mal-4(Ts)tetr

 strain D hisB24 leu-23

 

 (a) glucose minimal medium at 30°C

 (B) maltose minimal medium containing histidine at 42°C

 © complex medium containing tetracycline at 42°C

 (d) maltose minimal medium containing histidine and leucine at 30°C

 

2.The strain BL21(DE3) is commonly used for overexpressing proteins, either for enzymology or for crystallography or NMR. I need to Write the genotype of this strain. What carbon sources will it not grow on? Is it auxotrophic? Why are the lon and ompT mutations useful in this strain?

 

3. The strain XL1Blue is commonly used for cloning.I need to write the genotype of this strain. What carbon sources will it not grow on? Is it auxotrophic?
