Hi everyone, I'm interested in doing a yeast-2-hybrid on a very big protein (about 200 kDa large). Ideally, I'd like to use the full length cDNA as a bait, but this is probably not possible. Therefore, I was curious what I should do? If I used the full length cDNA, transform it into yeast, and I see expression on a Western Blot and growth on the SD/-Trp (Clontech Y2H kit), should I proceed to use the full length? Otherwise, what is the maximum size a bait can have?
