mRNA electroporation followed by microRNA transfection

hello every body



I am working mRNA delivery. as a part of the job, I have to try transfecting cells with an Invitrotranscribed mRNA containing target sequences for miR-143. here is my question:

1- Since I am transfecting cells with mRNA and it has a very transient nature, shall I co- transfect mature microRNA ( double stranded from mirvana mimics ,Ambion) at the same time or I'd better first transfect the mRNA and then microrna?

2- what is the best method for transfection of mammlian cells with mRNA?
