Hi all,

I plan to generate a new transgenic mouse where gene expression is prevented by upstream floxed STOP cassette:

-promoter-loxP-STOP-loxP-transgene-

Upon crossing to a CRE-recombinase expressing mouse line the STOP will be removed, leading to transgene expression.

Does anyone know of nay studies demonstrating the efficiency of different STOP sequences/cassettes? There are at least a couple of them at Addgene and apparently some more in Pubmed without any detailed information on the construction principles or even exact sequences.
