Linearising Cq values for Normfinder

Hey everybody,

I am trying out the Normfinder Macro for Excel.

In the FAQ it says to do this with Raw Cq values: "For assays amplifying at ~100% efficiency the Ct values can be linearized by Posted Image."

Does that mean the following in excel? =2^(-25.45)

I compared what I get from that formula with the example bladder cancer file they provide, but seem to get way smaller numbers for the input.

Or do I have to do =2^(-25.45+"Cq of GOI")? (GOI=gene of interest)
