I am using human monocytes/macrophages. Before performing flow cytometry, I block cells with 2% FBS in PBS solution. Should I use commercial Fc blocker to get rid of nonspecific antibody binding? Secondly, I am interested to see CD16, CD32 and CD64 expression. In that cases, should I block my cells? Yes, I am using isotype control in my flow panel.
