Hey all,

I recently took over the honcho position for a college level general bio lab. Unfortunately, many of the protocols were not left for me so I have been figuring it out on my own. I have reached a point, however, that I must reach out for help!

We have a lab that covers DNA and electrophoresis in which the students load dyes into a gel and run it. The dyes used are bromophenol blue (travels towards Cathode as DNA would) and crystal violet (should travel towards anode). It's not.

Here's the recipe I have tried:

.25g crystal violet
40g sucrose
volume to 100mL in 1X TBE

When this is run, it travels both directions (positive and negative?)!
I have tried different electrophoresis chambers and even bought new crystal violet. Does anyone have any experience with using this for teaching purposes? Does anyone have a different recipe or any ideas of possible problems?

Any help would be greatly appreciated!
