Is anyone else using AggreWell plates to form embryoid bodies? After 24 hours of incubation in the AggreWells, I am seeing poor incorporation of my single cells into embryoid bodies. I don't see nice, spherical aggregates. I'm wondering if anyone has advice on improving the yield.

I am using AggreWell 800 plates purchased in February 2013. My cells are from WiCell's WA07 (H7) line of human embryonic stem cells. The cells are normally cultured on Matrigel using mTeSR1. The embryoid body formation medium I use is 10% FBS, 0.1 mM NEAA, 0.1 mM BME, and 10 uM ROCK inhibitor in DMEM.

Thank you!
