Does anyone has a good method to extract genomic DNA from gram positive bacteria? I now use lysozyme at 37C for 1 h then add 10%SDS and incubate at 65C until solution is clear and after phenol:chloroform is addded then mix slowly by using cutting pipette tip and swirl gently. But I can not obtain a good one after check by agarose gel. Thanks.
