Please help.

1)I am trying to RTPCR a specific gene fragments from a viral RNA (negative sense).This viral RNA has a universal 12 bp conserved in the total 8 segments of the total viral RNA. so I have used this 12 bp for my RT at 37C for 1 hr. The longer fragments I should get is more than 2 kb. Is there 37C incubation for 1 hor too short for me to get the over 2 kb fragment. Please advise.

2) I have checked the sequence for the 8 fragments of the viral RNA, it has the universal 12 bp following two to three bp different for 8 segments What is the best way to amplifier specifically these 8 segments. Please advice.
